In this case the concentration of theophylline in a patient's body decreased from m to m in 24 hours and further in another 12 hours the drug concentration was found to m this indicate that in first 24 hours the drug had an effect on the body metabolism which the body became tolerant for the the next 12 hours. Therefore the rate constant in this case will become half for the metabolism of drug in the body.
